Tangential Flow Filtration System (TFF)

Sanitech Engineers is one of the reliable manufactures and suppliers of the latest cassette based tangential flow filtration (TFF) system meeting international regulatory standards with versatility of using it in multiple unit operations for volume reduction and purifying proteins, biomolecules, and other macromolecules. It is also recognized as one of the most trusted and versatile manufacturers in the industry.

As the industries work in set areas of applications, each has its own purity and quality to meet the demand. Some products are of very high value and low volume like products from biopharmaceuticals and others are high volume and low value like nutraceuticals. Thus, the membrane process system needs to be design such that the holdup should be less for biopharmaceutical/pharmaceutical products.

The system design is very compact and modular with ease of scalability, adaptability, and seamless integration into various process configurations.

FAQs

Tangential Flow Filtration (TFF) plays a role in the separation, purification, and concentration of therapeutic biomolecules. This method works by having the feed solution move along the membrane surface delaying the layer formation on the membrane surface.

It is a membrane filtration technique where the liquid flows parallel (tangential) to the membrane surface, instead of directly through it. This prevents clogging, allows continuous circulation, and enables the separation, concentration, or purification of biomolecules like proteins, peptides, and vaccines with high efficiency.

A TFF skid is a complete, skid-mounted system designed for tangential flow filtration. It integrates pumps, membranes, piping, sensors, and controls on a compact frame, allowing efficient concentration, diafiltration, or clarification of biomolecules. These skids are widely used in biopharma and peptide manufacturing because they offer scalability, automation, and reproducible results from lab to production scale.

The working principle of Tangential Flow Filtration (TFF) is that the liquid sample flows parallel (tangential) to the membrane surface, not directly through it. This sweeping action prevents clogging and allows continuous separation: Permeate (small molecules, salts, solvents) passes through the membrane. Retentate (proteins, cells, or larger biomolecules) is retained and concentrated. This principle makes TFF highly efficient for protein concentration, buffer exchange, and purification compared to dead-end filtration.

TFF filters come in four main types: flat sheet cassettes for lab and pilot use, hollow fiber filters for gentle handling of cells and viruses, spiral wound filters for cost-effective large-scale processing, and ceramic filters for harsh chemical or high-temperature conditions. The choice depends on the product’s sensitivity, process scale, and operating environment.

TFF is widely used in biopharmaceuticals for concentrating and diafiltering proteins, monoclonal antibodies, and vaccines. It is also applied in food, dairy, and nutraceuticals for clarifying juices, concentrating enzymes, and purifying probiotics, ensuring high yield with consistent quality.
